If reading for hours on end on a computer screen works for you then you probably don't need eink.
I have to say one reason to buy a Kindle over a Nook is Kindle customer service. It is really great. Any problem at all and they next day deliver a new one and send a mailing slip to send the old one back for free. They will help you figure out how to do things as well. They are great. B&N not so much from what I hear.

There have been very good answers from many others but I need to warn folks that the Kindle screen is not easier to read for everyone. The original reason I got an iPad was to read books as both paper and a more paper like Kindle are hard for me to read when I am tired. I have a much easier time on my MacBook so I knew the iPad would work better for me. So adding to the other answers, go to a store (hopefully near you) and try both not only for reading experience but other features as well.
