I cleaned up my computer today using Grand Perspective and I was able to either delete or move about 100Gs of files. As I suspected it's my video and slideshow files that are taking up lots of space. Also, my iPhoto library, although I didn't delete any photos from that yet.
Thanks for the great tip about Grand Perspective. It was extremely useful to detecting large files and also duplicate files....
